چکیده

Millipore diffusion chambers (MDC) with 0.22-j,m filters loaded with spores or vegetative cells of Clostridium botulinum were surgically implanted intraperitoneally (ip) into guinea pigs. MDC expose C. botulinum spores or vegetative cells to body fluids yet protect them from phagocytes. Guinea pigs receiving MDC containing 109 spores plus 108 polymorphonuclear (PMN) leukocytes and MDC with 109 vegetative cells died within 48 hr, indicating that toxin was released and diffused out. MDC with 109 spores alone allowed 90% of animals to survive for at least 96 hr. Microscopically, it was observed that vegetative cells in MDC were disintegrated and leukocytes plus spores were phagocytized and germinated; spores alone remained intact and phase bright. Chemotactic attraction of leukocytes to MDC walls was also observed. Apparently, body fluids do not attack spores; thus, PMN leukocyte engulfment is essential for germination and release of sporebound toxin in this type of C. botulinum pathogenesis. However, vegetative cells appear to be attacked by bacteriolytic enzymes (e.g., lysozyme) in body fluids, and leukocyte engulfment is not essential for toxin release.

Effects of T-2 Toxin on Cytokine Produc-tion by Mice Peritoneal Macrophages and Lymph Node T-Cells

Background: T-2 toxin is a mycotoxin of type A trichothecenes produced by several fungal genera such as Fusarium species. Mycotoxins can affect both cell mediated and humoral immune compartments. Objective: The purpose of this study was to investi-gate the effect of T-2 toxin on cytokine production by mouse peritoneal macrophages and lymph node T cells. Methods: Mouse peritoneal macrophages and...

Use of immunofluorescence and animal tests to detect growth and toxin production by Clostridum botulinum type E in food.

The appearance of Clostridium botulinum type E organisms and of toxin in experimentally inoculated packages of turkey roll was followed to study the time relationship between the presence of vegetative cells and the demonstration of toxin. The presence of vegetative cells was determined by immunofluorescence, and animal tests were used to assay toxin production. Growth initiated from detoxified...

Clostridium tetani growth and toxin production in the intestines of germfree rats.

Germfree rats were challenged orally and intrarectally with spores of Clostridium tetani. Although C. tetani spores remained viable in the intestinal tract, they were unable to germinate. Germfree rats were then challenged orally with vegetative cells of C. tetani. Vegetative cells were able to colonize the intestinal tract, replicate, and produce toxin. Tetanus antitoxin, but no tetanus toxin,...

Analysis of mosquito larvicidal potential exhibited by vegetative cells of Bacillus thuringiensis subsp. israelensis.

Vegetative Bacillus thuringiensis subsp. israelensis cells (6 X 10(5)/ml) achieved 100% mortality of Aedes aegypti larvae within 24 h. This larvicidal potential was localized within the cells; the cell-free supernatants did not kill mosquito larvae. However, they did contain a heat-labile hemolysin which was immunologically distinct from the general cytolytic (hemolytic) factor released during ...
